Background: Hydroxyurea is administered to control elevated blood counts in patients with myeloproliferative neoplasms (MPNs) reduce the risk of thrombosis and mortality. We reviewed side effects hydroxyurea, development secondary malignancies, drug resistance observed some patients.Current Concepts: The low incidences malformed births, intrauterine deaths, spontaneous abortions associated hydroxyurea therapy suggest that adverse pregnancy fetal are unlikely. However, animal studies have reported malformations; therefore, use during not recommended. Low-grade events include gastrointestinal symptoms such as nausea, upper discomfort, diarrhea. These usually resolved by dose adjustment or temporary discontinuation. Approximately 10% MPNs resistant another unable tolerate it owing effects. Drug intolerance most common causes inadequate cytoreductive MPNs.
